Tryfonia is L-5-hydroxytryptophan (5-HTP) derived from Griffonia simplicifolia seed extract. 5-HTP is a metabolite of the amino acid tryptophan, and an essential precursor of the biosynthesis of the neurotransmitter serotonin and the neurohormone melatonin.
Research supports a role for 5-HTP in supporting mood balance, sleep, and the proper tone of the muscles on the back of the neck with resulting changes in blood flow and pain fibers. Over 19 studies, conducted mainly in Europe, have evaluated the clinical effects of 5HTP.

A Swiss study found that a 300mg daily dose was slightly superior to fluvoxamine, a standard selective serotonin reuptake inhibitor (SSRI) with lower incidence of side-effects. 5-HTP has also been compared to tricyclic antidepressants (clomipramine and imipramine) and has been found to be as effective as these prescriptive antidepressants. Another advantage of 5-HTP is the quicker onset of action compared with conventional treatments. E.g. in one study, 50% of the patients began to improve after 3 days, and 80% within 2 weeks, whereas conventional antidepressants take much longer to achieve therapeutic response.
A Norwegian study found that 5-HTP improved sleep as effectively as L-Tryptophan. Melatonin is a down-stream metabolite of 5-HTP, which explains this effect. Studies in Spain and Italy found that 5-HTP was comparable to the drug methysergide in alleviating the symptoms of migraines (over 70% in both groups) while eliciting considerably fewer side effects.
Fibromyalgia patients have been found to have low serotonin levels and have shown symptomatic improvement with the use of tricyclic and SSRI antidepressants. Several clinical trials have confirmed significant improvement in symptoms, including pain, morning stiffness, anxiety, and fatigue. An Italian study showed significant improvement following a total dose of 300mg for 30 days. A longer 12-month study using a daily dose of 400mg of 5-HTP found the supplement to be as effective as amitriptyline and/or phenelzine (MAOI). No patient withdrew from the study.

Adult Dosage (MAX): Take one capsule three times daily with food, or as directed by a qualified health practitioner.
WARNINGS AND PRECAUTIONS:
To minimize gastrointestinal side effect risk, slowly increase dose over a 2-week period. Use for a minimum of 1 week to see beneficial effects for mood balance and 2 weeks for the alleviation of fibromyalgia symptoms. Consult a health care practitioner if symptoms persist and for usage beyond 1 year. Consult a health care practitioner prior to use if taking carbidopa or drugs/supplements with serotonergic activity. These may include, but are not limited to, L-tryptophan, Sadenosylmethionine (SAMe), St. John’s Wort, antidepressants, pain killers, cough/cold medication containing dextromethorphan, anti-nausea medication and anti-migraine medication. Discontinue use and consult a health care practitioner if you show signs of weakness, oral ulcers, or abdominal pain accompanied by severe muscle pain or if you experience skin changes. Some people may experience gastrointestinal disturbances such as diarrhea, nausea, vomiting and abdominal pain as well as drowsiness. Do not use if you have scleroderma. Exercise caution if driving or operating heavy machinery or engaging in activities requiring mental alertness. Do not take 5-HTP if pregnant, nursing, or trying to conceive. Not to be used by individuals under the age of 18 or those with a medical condition.
